Research Interests
The Inflammasomes and Cell Death Laboratory (INFeCT) investigates the molecular mechanisms of inflammasome complexes and Gasdermin proteins in the context of different pathologies.
Inflammasomes are cytosolic protein complexes that recognize or sense pathogen-associated molecular patterns (PAMPs) and danger-associated molecular patterns (DAMPs). Activation of the inflammasomes leads to not only the secretion of the pro-inflammatory cytokines IL-1beta and IL-18 but also triggers a Gasdermin-dependent inflammatory cell death called pyroptosis. Our research group aims to understand the molecular mechanisms of inflammasome regulation during infection, cancer and inflammatory diseases and to characterize new modulators of pyroptosis through the following projects:
- Characterization of new inflammasome and pyroptosis regulators
Since the overactivation of inflammasomes is deleterious for the host, identification of endogenously expressed proteins inhibiting inflammasomes is crucial. By using several molecular biology and genetic techniques, inflammasome and pyroptosis regulators are identified in bacterial and viral infections (TÜBİTAK1001, TÜBİTAK3501 project) - Elucidating immune escape mechanisms Host and pathogens develop a dynamic interaction with each other. Although our cells are equipped with receptors and sensors that detect and kill intracellular microorganisms, some of these pathogens develop sophisticated escape mechanisms to survive. Our group investigates different strategies bacteria employ to counteract innate immune proteins (ESCMID project).
- Evolutionary analysis of inflammasome- and Gasdermin- coding genes Inflammasome and Gasdermin genes are conserved throughout evolution in different species including bacteria. In an ongoing project, our group characterizes new Gasdermin-like bacterial proteins. Moreover, archaic variants from Neanderthals and Denisovans are functionally tested to gain insight into the evolution of innate immune genes (BAP and TÜBİTAK1002 projects).
